چکیده

Microarrays have unprecedented potential for the high-throughput detection and characterization of uncultivated microorganisms. Several different types of arrays have been developed or adapted for the interrogation of microbial genomes and monitoring microbial population dynamics and/or activity in relation to various microbial processes such as bioremediation and biogeochemical cycling. Identification and characterization of pigmented photoreceptor-producing microorganisms using FTIR spectroscopy

The identification, differentiation and classification of microorganisms have been subjects of research for many years. Recently, Fourier transform infrared (FTIR) spectroscopy techniques have gained attention in the characterization and classification of microorganisms based on biochemical profiles and cell structure characteristics. In the present study, the characterization and differentiati...

Isolating "uncultivable" microorganisms in pure culture in a simulated natural environment.

The majority (>99%) of microorganisms from the environment resist cultivation in the laboratory. Ribosomal RNA analysis suggests that uncultivated organisms are found in nearly every prokaryotic group, and several divisions have no known cultivable representatives. We designed a diffusion chamber that allowed the growth of previously uncultivated microorganisms in a simulated natural environmen...

Potential of DNA microarrays for developing parallel detection tools (PDTs) for microorganisms relevant to biodefense and related research needs.

Development of parallel detection tools using microarrays is critically reviewed in view of the need for screening multiple microorganisms in a single test. Potential research needs with respect to probe design and specificity, validation, sample concentration, selective target enrichment and amplification, and data analysis are discussed. Data illustrating selected probe design issues for dete...

Patterned Protein Microarrays for Bacterial Detection

Patterned microarrays of antibodies were fabricated and tested for their ability to bind targeted bacteria. These arrays were used in a series of bacterial immunoassays to detect E. coli 0157:H7 and Renibacterium Salmoninarum (RS). Microarrays were fabricated using microcontact printing (μCP) and characterized using scanning probe microscopy (SPM). The high-resolution SPM imaging showed that ta...

Identification and characterization of a novel fumarase gene by metagenome expression cloning from marine microorganisms

BACKGROUND Fumarase catalyzes the reversible hydration of fumarate to L-malate and is a key enzyme in the tricarboxylic acid (TCA) cycle and in amino acid metabolism. Fumarase is also used for the industrial production of L-malate from the substrate fumarate. Thermostable and high-activity fumarases from organisms that inhabit extreme environments may have great potential in industry, biotechno...
